VANGL1 overexpression enhanced this migration and wound closure in cultured cells. Downregulation of endogenous VANGL1 by small interfering RNA decreased the migratory response of epithelial cells to ITF stimulation.The deduced 524-amino acid protein has 4 transmembrane domains and a C-terminal ser/thr-x-val motif. STB2 shares 73.1% amino acid identity with STB1 (VANGL2) and 72.7% identity with Xenopus Stbm. Northern blot analysis detected 4.8- and 6.8-kb transcripts in nearly all human tissues examined.Northern blot analysis detected a 1.9-kb VANGL1 transcript abundantly expressed in testis and ovary, but not in any other tissue examined. Transfected cells expressed VANGL1 throughout the cytoplasm.Western blot analysis detected endogenous VANGL1 as a doublet of about 65 kD.
