PLCH2 is a member of the PLC-eta family of the phosphoinositide-specific phospholipase C (PLC) superfamily of enzymes that cleave PtdIns(4,5) P2 to generate second messengers inositol 1,4,5-trisphosphate and diacylglycerol.

By EST database analysis using Pseudomonas fluorescens phosphocholine-specific phospholipase C sequence as probe, Stewart et al. (2005) identified human PLCH2. The deduced 1,442 amino acid protein contains a pleckstrin homology (PH) domain, an EF-hand domain, a predicted Ca(2+)-requiring catalytic site containing X and Y domains, and a C2 domain. PLCH2 lacks a signal peptide. In comparison to phospholipase C delta-1 (PLCD1), PLCH2 contains conserved residues known to be involved in substrate binding and catalysis.
